Chemistry: keratin, bonds, pigment

A lash is a protein fibre held together by three kinds of bond. Water opens two of them. Only chemistry opens the third, which is what a lash lift does. Knowing which is which explains almost everything a lash will and will not tolerate.

Keratin

A ladder of fibres inside fibres

A hair is mostly protein, between 65 and 95 percent of its weight, and most of that protein is keratin. Humans have 54 working keratin genes, and 17 of them make hair keratins.42

Two keratin chains twist around each other into a coiled coil. Bundles of these form filaments about 8 millionths of a millimetre wide, the filaments gather into larger fibrils, and those fill the cells of the cortex, which carries about 90 percent of the fibre's weight and its strength. More than 80 smaller proteins lie between the filaments and cross-link them into one network.143

Almost all of this was measured on scalp hair. When eyelashes from 36 women were imaged with synchrotron X-rays, their inner build matched scalp hair closely.18

Bonds

One strong bond, two weak ones

Neighbouring keratin chains are tied together in three ways. Sulphur bridges, or disulphide bonds, are covalent and strong. Hydrogen bonds and salt links are weak.47

Water breaks most of the weak ones. Wet scalp hair needed about four times less force to start stretching, and broke at about a third less. Heat helps the weak bonds move too: dry hair softens at 144 degrees, but at ordinary room humidity a change shows near 55.789

This is why a heated curler or a wet lash only changes shape for a while. The weak bonds settle in the new position and let go again the next time the lash is wet.4

In the imaging study, women who wore mascara more often had more cracking in the outer layer of their lashes. Use of a mechanical curler showed no measurable effect. It was a correlation in 36 women, not a test.18

At the atelier

Curling a lash with heat or a curler is a hydrogen-bond trick, and it ends at the next wash. A lift is a different thing entirely, which is why we treat it as chemistry and not as styling.

The three bonds

Covalent and strong. Opened only by chemistry, as in a perm or a lash lift.
Hydrogen bond
Weak. Opened by water, re-formed as the lash dries.
Salt link
Weak. Opened by water and by strong acid or alkali.
Wet against dry
About a third less force to break a wet fibre.

Water opens the weak bonds. Only chemistry opens the strong ones.

That is why a curler's shape lasts until the next wash, and a lift stays in the lash it was set in.

Surface

A water-repellent skin, and what alkali does to it

The outside of the cuticle carries a branched fatty acid called 18-MEA, bound directly to the protein. It is what makes a healthy fibre shed water. Strip it and the fibre turns water-loving, and alkaline treatments are what strip it.6

In liquid water a hair swells by 10 to 15 percent in width and hardly at all in length.5

The surface carries a charge that flips at an acidic point, reported between pH 2.5 and 4.5. Between pH 7 and 10 a fibre takes up little alkali. Above 10 it swells, its salt links break and alkali begins to cut its bonds for good.327

Strength

How much a hair can take

Pulled until it breaks, a hair holds 150 to 270 million pascals, depending on humidity and speed. Measured on fibres from 13 donors, it behaves like a spring up to about 3 percent stretch, then yields and flows, then stiffens again, and stretched as much as 50 percent before breaking.1011

The long flat part is thought to be the spirals of keratin unwinding. A wet hair can be stretched about 30 percent without lasting harm.105

No one has measured an eyelash this way. Every figure here comes from scalp hair, and the same is true of almost every claim made about what treatments do to lashes.

Pigment

Two melanins and a late grey

Lash and hair colour come from two pigments: brown to black eumelanin, and yellow to reddish-brown pheomelanin, which contains sulphur. Both start from the amino acid tyrosine and the enzyme tyrosinase; adding cysteine tips production toward the lighter one.13

Black, brown and blond hair differ mainly in how much eumelanin they hold. Only red hair carries the two in about equal amounts. The pigment sits in the cortex, never the cuticle, and is made only while the hair is growing.12414

By middle age, three people in four have some grey scalp hair. Lashes grey far later. Their pigment cells carry an enzyme that scalp pigment cells lack. That this is the reason is an idea, not a finding.151716

Pale or greying lashes often look sparse when they are not. A tint, where it suits, or a soft brown or black fibre can show what is already there.

Lash lift

A permanent wave on a ten millimetre fibre

A lash lift is the same chemistry as a hair perm. An alkaline lotion opens the cuticle so a reducer, usually a thioglycolate, can reach the cortex and break the sulphur bridges. The lash is held on a shield in its new shape, and an oxidiser joins the bridges again in their new positions.2320

The law treats it as chemistry. In the European Union and in Canada, products for curling eyelashes may contain at most 11 percent thioglycolic acid, at pH 9.5 or lower, for professional use only. The European safety opinion behind that limit rested on one in-use test with ten people.252824

What it does to a fibre was measured on scalp hair. After a full perm most sulphur bridges had re-formed, but the order of the protein was disturbed and the fibre was weaker, with some cysteine oxidised past the point of re-joining. How many bridges break and re-form is still called the main gap in the field. No study has done this on lashes, or on repeated lifts. Cysteamine, the reducer in so-called gentle perms, is itself a recognised contact allergen.21221930

Sulphur bridges hold the two chains in their natural shape.

We work within the Canadian limits and only on lashes that are healthy enough for it. We will not tell you a lift is harmless or quote a safe interval between lifts: that has never been measured. If your lashes need a rest, we say so.

Tint

What a tint is, and what the law allows near the eye

A permanent dye starts colourless. Small molecules such as p-phenylenediamine (PPD) soak into the swollen fibre with an oxidiser and join into large coloured molecules that stay trapped inside.23

PPD is a known allergen: among dermatitis patients who are patch tested, about 4 to 6 percent react to it. In published reports of harm from lash dyeing, allergic dermatitis was the most common outcome.2331

Three regulators draw three different lines. Canada does not permit coal-tar dyes such as PPD in the area of the eye at all. The European Union allows them on lashes for professionals at up to 2 percent. The United States lists one colour additive for lashes and brows: silver nitrate, applied by a professional for no more than a minute.28262729

What the evidence shows

A keratin lift puts keratin back into the lash.

No evidence was found that keratin applied to the surface enters or rebuilds a lash.

A lift breaks 20 to 30 percent of the bonds and the neutraliser re-forms 90 percent.

No measurement of this was found, for scalp hair or for lashes.19

Hair has a pH of 4.5 to 5.5.

A solid fibre has no pH. What is measured is the point at which its surface charge flips, between 2.5 and 4.5.3233

No colour additive is approved for eyelashes in the United States.

Out of date since a 2021 rule listed silver nitrate for professional lash and brow colouring.29
